Berry Full Of DNA

Materials:
1) Take the fruit and cut it into small pieces and put the pieces into a plastic bag
2) Add 20 ml of the shampoo/salt solution to the bag and squeeze the bag to remove all excess air
3) Being careful not to break the bag, mush the fruit pieces for 5 minutes
4) Put the bag on ice for 1 minute
5) Mush the fruit solution again for 3 minutes
6) Put the fruit mixture back on ice for an additional 1 minute
7) Make a funnel out of the coffee filter and pour the slurry into the funnel which is on top of the cup/beaker
8) Keep the filtered solution and put about 3 ml into a 20 mL test tube.
9) Add 2 ml of the 95% ethanol to the tube by drizzling it down the side of the tube. DO NOT SHAKE THE TUBE!!!!
10) Let the tube stand in rack for 5 minutes
11) Look at what is happening at the interface of the ethanol and the fruit extract- you should see translucent DNA between the layers.
12) Take a glass rod or hook and carefully move it clockwise to spool out the DNA.
- Fruit

- Shampoo/Salt solution (20 ml per group)- 20% shampoo and 1.5 M NaCI
- Ice in cooler
- #6 coffee filters
- Beakers
- Funnels
- 20 ml test tubes- one per group
- 95% ethanol- 200 ml
- Small tubes for clean DNA
